Biotech for All

- Research extends from spec. labs to kitchens, workshops and DIY labs - Democratisation -> threat and opportunity

- 1990, software dev and biotech started DIY and open source strats by experts, uni, large companies to broader community - 2008, tech enthusiasts in Boston took biotech research from institutions to garages and kitchens in urban/rural. - eBay: garage labs with scales, mixers, refrigerators, incubators - laymen and researchers together for biotech or DIY satisfaction

Microscopes form cheap webcams:

- parallel dev of DIY exists tech components for dev homemade bioanalysis equipments i.e., microchips, LEDs have become cheap for laymen - spectrometers, microscopes, dna seq machinery

- hacking part of: cheap, state-of-the-art for mass market are modified for new functions with lab capabilities - internet: build computer comp uscope from webcam - electronics: unchanged. position of lens is altered - solid platform fixes object of study, illuminated by LED

- Advancement of DIY Bio due to Web 2.0 and social media - founded labs were networked through blogs for everyone with internet connections instead confined to journals - new discoveries shared via internet - copyleft > copyright - DIY bio is expression of new, contemporary desire for community DIY.

New Dangers and limits:

- USA: debate - hiding places for bio war agents/accidents - FBI courses for garage labs directors and trains to report suspects early

- analytical activities including service sector, labs required to comply with national/international rules and std. for choice methods and instruments - labs have to accredited -> areas will be reserved for professionals and speci. labs

Potential for developing countries and teaching:

- DIY should not be primitive, instead pursued in line with scientific ethics and std. - diy bio suggest infrastructure not enough for producing danger also no interest as society - potential: underdeveloped countries DIY offers alternatives - 3 inventors from Netherlands: hairdryer, shoebox and electronics to develop malaria detector, now used in Africa - less expensive than PCR (duplication method for nucleic acids using PCR to quantiy DNA)

- opportunities in teaching - ETH Prof Gerd Folker: meaning of grasp: understanding and grasp wrongly used synonyms - grasp: using mind and body to interpret object: we need this to form picture of surroundings - understanding: mental process - grasp = garage labs and DIY: Philosophy - instead of expert -> learner, rather people can bring own knowledge

DIY in Bioanalytics: Doing and grasping it yourself

- Oct 2014, FHNW and Hackteria, SATW organised 2 day workshop - Goal: Biohacking and DIY strats used as teaching units

- 7 workshop participants making temp. biotech lab

Phosphatase activity text with potatoes:

- first thematic portion: crash course in electronics and asked to perform experiments using LEDs, button cells, resistors, multimeters - followed by kitchen lab learning natural enzyme isolation and concentration - peeled & chopped potatoes, pressed with garlic press, separated liquid from solid using filter paper - pipetting, potato extract used to create dilution series enabling phosphate activity test with para nitrophenyl phosphate - phosphatases catalyse colourless substance breakdown into para nitrophenol (yellow, measurable its concentraion spectroscopically 405 nm) - using quantity para nitrophenyl phosphate added and quantity para nitrophenol measured, determinably of phosphate activity

- technical portion: rapid prototyping: DIY measuring equipments to analyse dilution series - discussion of designs and everyone assembled personal measuring instrument - aim: trial and error experiment. create stable measurement setup consisting of cuvette holder, stray light protection, power supply and measurement electronics -> used by DIY biologists to measure phosphate activity in potato extract

- for spectrometry principle and calibration, the measurement setup combined with theoretical elements to translate results - intro to beer-lambert law on attentuation light radiation intensity acc. to material absorbed - experienced ones, used to ucontrollers arduino to convert setup enabling display and analyse reulsts from hommade spectro on laptop - controller linked via usb using open source software from internet

Insight into the black box:

- end of second workshop day, discussion of results - summary: some wanted theory, some hands on - lack of clear instructions and tools -> boosted creativity

Videos

Greg Gage Ted
